Forest decline is a complex process involving interactions between environmental stressors, host condition, and associated microbial communities. This study investigated the microbial composition of Scots pine trees exhibiting decline symptoms in a forest near Sopron, Hungary, and compared them with neighbouring non-symptomatic individuals. In addition, remote sensing data based on Normalised Difference Vegetation Index (NDVI) were used to assess vegetation dynamics within the study area. Microbial communities were analysed using both shotgun metagenomic and 16S rRNA amplicon sequencing approaches. The results showed that community composition was primarily structured by tissue type rather than tree health status. Soil samples exhibited the highest diversity, while needle and trunk-associated communities were more constrained. Symptomatic samples showed a decrease in taxon richness, although overall community composition remained relatively stable. Specific taxa, such as members of the Acidobacteria phylum, were reduced in symptomatic samples, while some opportunistic and low-abundance taxa showed slight increases. Remote sensing analysis revealed a clear reduction in NDVI values around 2021, indicating decreased vegetation vigour. This pattern was consistent across multiple datasets and supported by spatial and temporal analyses. The combined results suggest that tree decline in the study area is associated with subtle microbial shifts and measurable changes in vegetation condition. These findings highlight the potential of integrating microbiome analysis with remote sensing approaches to better understand forest health dynamics.
